During my M.S. thesis work I was involved in amultistep synthesis of solid support bearing amodified nucleoside for automated synthesis ofsiRNAs. First, I developed a three-step synthesis ofprotected 5'-amino uridine from uridine. Next Iprepared the 3'-succinate of 5'-amino uridine in aone-pot two-step synthesis and attached it to a solidsupport via a succinamide linker. I had analyzed andcharacterized all new compounds using NMR techniques.Others in our laboratory used this solid support forsynthesis of amide-modified siRNAs. We anticipatethat the amide linkages will facilitate crossing thecellular membrane and improve nuclease resistance,biodistribution and pharmacokinetics of modified siRNAs.

Suresh SwapnaSwapna Suresh has graduated with a M.S. in Chemistry fromNortheastern University, Boston, Massachusetts, U.S.A. In May2007, she joined Prof. Rozners' group and her thesis was focussedon the synthesis of 5'-amino protected uridine and its loading onthe solid support LCAA-CPG . The project wasfinished by June 2008.
